Spywareblaster runs in real-time, Superantispyware (free version) doesn't, so they won't bother each other.
Having couple of antimalware programs handy is always a good idea.
They are very different beasts and I would run them together.
Superantispyware is a true antispyware with malware definitions and cleanup routines built into the program. You should update it and perform manual scans with it on a regular basis.
SpywareBlaster sets block lists of known bad sites/cookies for Internet Explorer and known bad cookies for Firefox. These settings are passive and all you need to do is update it regularly and make sure all the protection is enabled.
